Polycyclic aromatic hydrocarbons (PAHs) are a typical class of persistent organic pollutants that is ubiquitous worldwide. Previous animal studies suggested that PAHs had adverse effects on female reproduction. However, the human data regarding relationship of PAHs exposure with women reproductive health, such as ovarian dysfunction, are scarce. In this case-control study, the associations of serum levels of PAHs with the risk of premature ovarian failure (POF) and reproductive hormones in Chinese women were investigated, with recruiting 157 POF patients and 217 healthy women. The serum levels of 12 types of PAHs, as well as reproductive hormones, including follicle-stimulating hormone, luteinizing hormone and anti-mullerian hormone, were determined. In the logistic regression models, most individual PAH congeners showed significantly positive correlations with the risk of POF (p < 0.05), except for fluorine and pyrene. Benzo(a)pyrene (BaP), as the most carcinogenic PAH congener, was observed to be significantly positively associated with the risk of POF. After adjustment for age, body mass index, educational levels and household income, per one-unit increase in the log-transformed BaP concentration was significantly correlated with 2.191-fold increased risk of POF (OR = 2.191, 95%CI: 1.634–2.938, p < 0.05). To the best of our knowledge, this is the first study to report an association between internal exposure levels of PAHs and the increased risk of POF in women. 相似文献

In the present study, groups of juvenile Atlantic salmon (Salmo salar) were fed gelatine capsules containing fish-food spiked with PFOA or PFOS (0.2 mg kg−1 fish) and solvent (methanol). The capsules were given at days 0, 3 and 6. Blood, liver and whole kidney samples were collected prior to exposure (no solvent control), and at days 2, 5, 8 and 14 after exposure (Note: that day 14 after exposure is equal to 7 d recovery period). We report on the differences in the tissue bioaccumulation patterns of PFOS and PFOA, in addition to tissue and compound differences in modulation pattern of biotransformation enzyme genes. We observed that the level of PFOS and PFOA increased in the blood, liver and kidney during the exposure period. Different PFOS and PFOA bioaccumulation patterns were observed in the kidney and liver during exposure- and after the recovery periods. Particularly, after the recovery period, PFOA levels in the kidney and liver tissues were almost at the control level. On the contrary, PFOS maintained an increase with tissue-specific differences, showing a higher bioaccumulation potential (also in the blood), compared with PFOA. While PFOS and PFOA produced an apparent time-dependent increase in kidney CYP3A, CYP1A1 and GST expression, similar effects were only temporary in the liver, significantly increasing at sampling day 2. PFOA and PFOS exposure resulted in significant decreases in plasma estrone, testosterone and cortisol levels at sampling day 2, and their effects differed with 17α-methyltestostrerone showing significant decrease by PFOA (also for cholesterol) and increase by PFOS. PFOA significantly increased estrone and testosterone, and no effects were observed for cortisol, 17α-methyltestosterone and cholesterol at sampling day 5. Overall, the changes in plasma steroid hormone levels parallel changes in CYP3A mRNA levels. Given that there are no known studies that have demonstrated such tissue differences in bioaccumulation patterns with associated differences in toxicological responses in any fish species or lower vertebrate, the present findings provide some potential insights and basis for a better understanding of the possible mechanisms of PFCs toxicity that need to be studied in more detail. 相似文献

In order to study the time-dependent effects of chlordane on thyroid hormone levels and histological structure in rats, 120 healthy 4-week-old SD rats were divided randomly into 12 groups. Rats in treatment groups were treated orally by gavage with chlordane at a dose of 15 mg kg?1 for 5, 10, 15, 20, 30, or 40 days, while rats in control group received an equal volume of corn oil. Serum levels of thyroxine (FT4), triiodothyronine (FT3), and thyroid stimulating hormone (TSH) were measured by radioimmunoassay. Sections of thyroids were dyed with hematoxylin–eosin to detect any pathological alterations. In chlordane treated groups, on day 5, changes in serum levels of FT4, FT3, and TSH were not found. On days 10 and 15, serum levels of FT4 and FT3 decreased, accompanied by increased levels of TSH. On days 20, 30 and 40, serum levels of these hormones returned to control values. As for alterations in histology, on day 10–15, follicular epithelium of the thyroid were found to proliferate into two layers in chlordane treated rats. On day 20, in focal areas the two-layer afollicular epithelia were observed to continuously proliferate into 3–4 layers of follicular epithelium, termed hyperplasia plaques. On days 30 and 40, solid buds and secondary follicles to solid buds were detected. The earliest alteration of thyroid hormones in treated SD rats occurred between 5 and 10 days with corresponding histopathologic changes. 多溴联苯醚(polybrominated diphenyl ethers,PBDEs)是一类常用的溴代阻燃剂,由于使用广泛,已成为普遍存在的环境污染物。研究表明,PBDEs具有内分泌干扰作用,影响甲状腺功能,对胚胎发育、婴幼儿及儿童的健康存在潜在的危害。本研究采集了100例具羊水穿刺指征的孕妇的孕中期羊水样本,采用罗氏全自动电化学发光免疫分析仪测定羊水中促甲状腺激素(TSH)、三碘甲状腺原氨酸(TT3)、游离三碘甲状腺原氨酸(FT3)、甲状腺素(TT4)、游离甲状腺素(FT4)、抗甲状腺过氧化酶抗体(TPOA)和抗甲状腺球蛋白抗体(TGA)的含量,用气相色谱-负化学源-质谱法测定羊水样品中16种PBDEs同系物含量。结果显示,羊水中TSH、TT3、FT3、TT4、FT4中位数分别为0.342μIU·m L-1、0.949 nmol·L-1、1.52 pmol·L-1、11.02 nmol·L-1和3.91pmol·L-1,95%参考区间值分别为0.174~0.872μIU·m L-1、0.835~1.01 nmol·L-1、1.26~2.72 pmol·L-1、9.42~20.68 nmol·L-1和2.16~6.35 pmol·L-1。具有唐氏筛查高风险和甲状腺激素水平低下的羊水样品中共检出6种BDEs(BDE-47、BDE-71、BDE-138、BDE-183、BDE-190、BDE-209),其中BDE-71、183和190在被检羊水样品中均被检出,并且,TSH水平低下的羊水中BDE-190和209负荷水平显著高于正常组。这些结果提示,大连地区孕中期妇女羊水存在PBEDs内暴露,并且PBDEs内暴露可能与羊水的甲状腺激素水平低下、尤其TSH水平升高有关,提示PBDEs暴露可能与胎儿中枢神经系统损伤产生的下丘脑-垂体-甲状腺(HPT)轴功能失调有关。因此,羊水PBDEs内暴露对新生儿出生结局的影响值得进一步关注。 相似文献

This study was undertaken to validate potential biomarkers of exposure and effects due to chemical contaminants in breedingcolonies of the Great Blue Heron and the Black-crowned Night-Heron on the St. Lawrence River. Eggs and fledglings from both species were collected from many colonies along theRiver. The fledglings from colonies in freshwater and brackishwater were more contaminated by mercury and PCBs than those from estuarine and gulf colonies. With respect to fledglings ofthe two heron species, some morphometric and blood biochemicalmeasurements, including plasma thyroid hormones and retinol, were significantly different among colonies. Significant differences were also observed in liver retinoids, EROD and porphyrins among colonies. The results of this study suggestthat plasma retinoids and thyroid hormones are good biomarkersof exposure and effects, and are sufficiently sensitive to reflect local and regional variations in contamination. Along with the measure of contaminants in egg and plasma, they constitute non-invasive biomarkers which represent an importantcriteria for long term monitoring of wildlife species. Lead (Pb) accumulation in Sesbania drummondii shoots was enhanced by 654 and 415% in the presence of 100 microM IAA and 100 microM NAA, respectively, compared to control plants (Pb alone). However, when IAA or NAA was added along with EDTA, Pb accumulation further increased in shoots by 1349% and 1252%, respectively. Scanning electron microscopic observations revealed that Pb particles in both leaf and stem of Pb+EDTA+IAA and Pb+EDTA+NAA treated plants were concentrated in the region of vascular bundles. In root tissue, Pb particles were present between epidermis to stele region. Plant growth in both treatments (Pb+100 microM IAA and Pb+100 microM NAA) was comparable to control plants; however, it was significantly inhibited in the treatments containing Pb+EDTA and EDTA at concentrations of 10 microM of IAA or NAA. Moreover, the photosynthetic efficiency and strength of the treated plants were not affected in the presence of IAA or NAA and EDTA. 相似文献

用不同剂量的2,3,7,8 四氯苯二英(2,3,7,8 tetrachlorodibenzopdioxin,TCDD)对小鼠胚胎附植数量和生长发育情况进行剂量反应评估,分析TCDD对假孕小鼠子宫蜕膜生长状况的影响以及比较胚胎种植前后染毒效果.所有样本采集血液,测定母体血清雌二醇和孕酮的浓度变化.结果表明,50和100ng·(kg·d)-1TCDD处理的孕鼠,胚胎附植数量明显减少(P<0 05),着床后胚胎出现发育迟缓,质量减轻(P<0 05);同等剂量TCDD处理的假孕小鼠,子宫蜕膜生长受到了极显著的抑制(P<0 01);种植前后染毒效果比较发现种植前期染毒的小鼠胚胎对TCDD更为敏感.母体血清雌二醇浓度随剂量和染毒时段有不同程度的升高,而孕酮浓度则极显著下降(P<0 01).由此表明低剂量TCDD可能通过对生殖激素的干扰,强烈影响NIH小鼠妊娠早期生殖生理状态和着床后胚胎的发育.实验同时证明小鼠也可作为早期妊娠毒物研究的供试动物,用以筛选动物和人类早期妊娠的有害化合物. 相似文献
